Exeter City manager Paul Tisdale says the search for his new No. 2 will begin immediately after Joe Gallen quit St James's Park less than three months into the job.
The former Queens Park Rangers head of youth has accepted a role in Millwall's youth academy.
Tisdale made it his priority during the summer to appoint a suitable successor to Paul Buckle, who left to become the new manager at Blue Square Premier rivals Torquay.
But after some of his prime targets rejected the post, including Team Bath boss Andy Tillson, the City boss turned to Gallen who was looking for a route back into football after being sacked by QPR.
His departure will come as a blow to Tisdale, who now faces a fresh search for a suitable assistant.
"It's a real shame because he had settled in really well," said Tisdale. "He is a very good coach and Joe was beginning to understand what I was looking for. But it means we will have to find someone else.
"There is going to be a period now when we are short in terms of the coaching side and it is a bit of a blow for us, to be honest, because I waited a long time to get someone who I considered to be the right person for the job.
"I went through the whole of pre-season without someone with me because I was waiting for that appointment and it is disappointing.
"But we will move on and we wish him all the best because he has been a good chap, good company and he has put a lot of effort in."
Tisdale is considering the possibility of appointing a short-term assistant whilst he searches for a permanent successor.
"We have got a board meeting on Monday and that is one of the things we need to discuss," he said.
"Steve (Perryman) and I need to have a discussion about it.
There is a chance that we might get someone in temporarily, just to get us through the next four or five weeks, before we actually make an appointment." Exeter Express and Echo

"The departure of assistant boss Joe Gallen will not derail Exeter City's FA Cup ambitions, according to Grecians manager Paul Tisdale.Gallen's sudden exit to Millwall came as a huge shock this week and another blow to a club desperately trying to inject some momentum into their season.
An injury crisis at the beginning of the season, a succession of frustrating draws on the pitch, suggestions that some of their players had been speaking to rival clubs and now Gallen's swift exit means it has hardly been an ideal start to the campaign.
But there's nothing like a good old FA Cup clash to get the pulses racing again and provide a springboard for a turnaround in fortunes.
Tisdale insists that the latest setback will not disrupt preparations ahead of City's trip to Stonebridge Road to face Blue Square Premier outfit Ebbsfleet United.
"I was surprised about what happened with Joe but that is football and you have to move on," said Tisdale.
"It is not going to affect us on Saturday. We might have someone different doing the warm-up but we are not going to be disrupted."
The defiant stance is understandable as Gallen's departure could not have come at a worse time.
Tisdale spent the summer searching for a successor to Paul Buckle, who quit St James's Park to take over at Devon rivals Torquay United.
In the end Gallen was invited to take the job on a trial basis while he sorted out his messy divorce from Queens Park Rangers. And while the search is already underway for a new assistant, Tisdale will spend this Saturday trying to navigate City's passage into the first round of the FA Cup. ......"
